Аннотация

Пренатальный стресс рассматривается в качестве фактора риска развития в последующей жизни таких заболеваний, как психические, сердечно-сосудистые и метаболические расстройства. Дисрегуляция активности гипоталамо-гипофизарно-адренокортикальной системы (ГГАС) служит одним из механизмов возникновения этих расстройств. Регуляция ГГАС осуществляется посредством двух типов рецепторов - минералокортикоидных (МР) и глюкокортикоидных рецепторов (ГР) кортико-лимбических структур мозга. Связь между пренатальным стрессом, полом и активностью ГГАС, а также роль центральных МР и ГР в этом взаимодействии исследованы недостаточно. Нами изучено влияние пренатального стресса на активность ГГАС взрослых самцов и самок крыс и экспрессию ГР и МР в гиппокампе и медиальной префронтальной коре (мПФК). Пренатальный стресс моделировали, подвергая беременных самок крыс линии Вистар одночасовой иммобилизации с 15-го по 19-й день беременности. У взрослых потомков изучена базальная активность ГГАС и стрессорный уровень кортикостерона в ответ на 30-минутную иммобилизацию. Экспрессию ГР и МР оценивали методом вестерн-блот и дополнительно в гиппокампе иммуногистохимическим методом. У пренатально стрессированных самцов выявлено пролонгирование стрессорного ответа, а у самок - повышение базальной и стрессорной реактивности и усилении чувствительности ГГАС к сигналам обратной связи. У самцов изменения активности ГГАС сопровождались снижением содержания ГР и МР в гиппокампе и мПФК при неизменном соотношении ГР:МР. У пренатально стрессированных самок наблюдалось усиление экспрессии белка ГР и снижение количества белка МР в гиппокампе и увеличение соотношения ГР:МР в обеих областях мозга. В гиппокампе самцов и самок наибольшие изменения содержания ГР и МР выявлено в СА3 поле и зубчатой извилине. Полученные данные указывают на вклад изменения экспрессии белков ГР и МР гиппокампа и мПФК в зависимую от пола модификацию активности ГГАС в результате пренатального стресса.
