Аннотация

Целью работы было изучение роли оксида азота (NO) и простагландинов (ПГ) в модулирующем действии слабой ирритации слизистой оболочки желудка (СОЖ) на желудочную секрецию, вызванную раздражением блуждающего нерва. У наркотизированных крыс в условиях полостной перфузии желудка секрецию кислоты и бикарбонатов рассчитывали на основе измеренных в перфузате значений pH/PCO2. Концентрация пепсиногена определялась по деградации гемоглобинового стандарта. Полостная перфузия желудка раствором слабых ирритантов (500 мM NaCl, pH 3.0) в сравнении с перфузией контрольным изотоническим раствором (154 мM NaCl, pH 4.0) усиливала секрецию H+ и HCO3-, вызванную электрическим раздражением периферического отрезка субдиафрагмального вагуса. Селективная блокада нейрональной синтазы оксида азота (nNOS) с помощью 7-нитроиндазола, а также неселективная блокада нейрональной и эндотелиальной NOS с помощью Nω-нитро-L-аргинина в равной мере подавляли потенцирующий эффект ирритации. Блокада циклооксигеназы индометацином не влияла на потенцирующий эффект ирритации, но подавляла вагусную секрецию H+ и HCO3- на фоне перфузии желудка контрольным изотоническим раствором. Вагусная продукция пепсиногена ослабевала в присутствии блокаторов NOS независимо от ирритации. Сделан вывод о том, что потенцирующий эффект ирритации СОЖ по отношению к вагусной секреции кислоты и бикарбонатов опосредуется локальной активацией nNOS, но не зависит от продукции ПГ.
